Dr. D'Amato has also been working to create a transgenic mouse that will express VEGF in the RPE cells beneath the retina to induce CNV. In this continuation of last year's experiments, he is utilizing a genetic element called a promoter that will cause the tissue-specific expression of the VEGF gene in the RPE cells. In addition, he plans to introduce into the same mouse line bacterial genetic elements that will allow for VEGF to be turned on and off in the presence and absence, respectively, of the antibiotic tetracycline. This inducible effect, if successful, will have the advantage of promoting CNV in older mice to more closely mimic the pathology of age-related macular degeneration (AMD) in humans. These mice could represent a more useful animal model for further study and testing.
